Abstract: Intracellular movement of diverse cargos to their destination compartments is a hallmark of mammalian cells. The complexity of the cellular structural components and regulators involved in the secretory pathways has garnered the interest of many cell biologists. However, till date, much remains inconclusive and debatable, especially regarding the machinery involved for transport from Golgi to cell surface. Here, the role of Rab6 and Mon2 proteins in formation and transport of carriers were investigated using budding and single particle analyses, respectively. I carefully chose the two constitutive cargo of interests – TNF-α and lysozymeC, which were found to co-localize within Rab6-associated post-Golgi carriers in preliminary studies. The effects which ensued the knocking out of Rab6 showed that it impedes movement of the cargo carriers, as well as reduces steady state cargo biogenesis. Future studies involving Mon2 and other secretory cargos should be performed to elucidate the significance of this Golgi protein in the dynamics of carrier transport.
