Abstract: Fluorescence microscopy is a technique that uses fluorescence to study the properties of samples. Fluorescence microscopy is rapidly evolving and is being widely used in biological areas. Thanks to recent development, light sheet fluorescence microscopy (LSFM), first introduced in the 20th century, is being developed as an efficient fluorescence microscopy technique. However, the image quality of conventional LSFM does not meet the requirements of modern research, as well as the decrease of contrast due to the show up of ghost shapes and background noise. The aim of this study is to introduce a Bessel beam and a polarization selective modality to reduce the background noise and improve the contrast of image. Firstly, a Bessel beam based LSFM system is built, and its lateral and axial resolution are measured. Secondly, a polarization selective modality is used to improve the image quality. With this Bessel beam based multimodality LSFM, the contrast of the image is enhanced. A 3D visualization of a mouse kidney section sample is reconstructed by the Bessel beam based LSFM with a selective polarization modality. Finally, the thesis also gives the potential methods and applications of LSFM with future work directions.
