The lack of extracellular amino acids can lead to the induction of the integrated stress response (ISR). The ISR is mediated by phosphorylated eIF2-alpha (eIF2α) and activating transcription factor 4 (ATF4), with the latter transactivating target genes in numerous pathways, including that of De Novo Serine Synthesis (DNSS).
