Abstract: Direct electron transfer (DET) of proteins has attracted much attention due to its important applications in bioelectronics and drug delivery system. This project studies nanostructure related DET behaviour of hemoglobin (Hb) and its biosensing. Different dimensional gold (Au) nanostructure, namely, zero-dimensional (0D) Au nanoparticles, 1D Au nanorods, 2D Au nanofilm and 3D Au ordered porous nanofilm, were fabricated and characterized. Bioactivity related conformation structure of Hb and the DET behaviours of Hb immobilized on Au nanostructures were investigated by FTIR and cyclic voltammetry respectively. Results demonstrate that 3D porous Au nanofilm produces the best DET behaviour of Hb, which results in highest sensitivity in the biosensing of H2O2. This project provides information in understanding the nanostructure-dependent DET of protein and its related applications in sensitive biosensing.
