|Title:||Study of static cell culture on a three dimensional (3D) scaffold||Authors:||Jebu Mathen Varghese||Keywords:||DRNTU::Engineering::Bioengineering||Issue Date:||2011||Abstract:||Tissue engineering has gained the interest of modern medicine so as to address the problem of the shortage of tissues and organs for transplantation and repair. One of the techniques employed today for engineering tissue is the seeding and culturing of cells on a three-dimensional biodegradable scaffolds in vitro. For successful tissue engineering results, it is necessary to select an appropriate scaffold and to optimize the cell seeding and cultivation in vitro. It is therefore necessary to study and analyze the responses of cells on a scaffold under different situations. This project was carried out in two phases. The first phase was to study of cell adhesion, growth and migration resulting from the seeding of different cell density of Human Fibroblast cells (HDF). The cells were statically seeded onto collagen scaffold of the same thickness. Four different cell densities were seeded onto these scaffolds. In phase two, the most efficient cell density based on the results of phase one, was seeded on to a new set of scaffolds of different thickness. Two different thicknesses of scaffolds were used, 4mm and 2mm. In both phases the cells were allowed to grow and migrate on these scaffolds for a period of fourteen days with culture medium changed every two days.
