Abstract: The RUNX3 gene is essential for the development of multiple cell lineages and is frequently involved in a variety of cancers and immunological diseases. Despite such frequent association with human diseases, the transcriptional regulation of Runx3 remains elusive. Owing to the huge ~350 kb Runx3 locus and complexity of the use of two alternate promoters for transcription, the search for regulatory regions outside the immediate promoters by traditional methods such as DNase I hypersensitive assay, proved to be a daunting task.
