Abstract: With the rapid development of enzyme biosensor, glucose oxidase, one of common but important enzyme in glucose determination, has been studying and applying in biosensor field for a long time. In order to improve the immobilization technology of glucose oxidase for biosensing, we decide to modify the TEOS sol gel process to prepare a glucose oxidase friendly carrier. In this paper, glucose oxidase(GOD) is immobilized in the TEOS sol gel with pH=5.5, which is prepared by combining the acid based sol and alkaline base sol, called two steps sol gel process. The type of chemical groups in this sol gel production and its impact on the activity of glucose oxidase are determined by the Fourier transform infrared spectrometer (FTIR) and ultraviolet and visible spectrophotometer (UV-VIS), respectively. It is demonstrated that the product prepared by this two steps method accords with Si02 structure, as there are three . absorbance peaks at 1086cm-l, 813cm-l, 477cm-l which are belong to Si-O-Si characteristic absorbance peale In addition, the activity of glucose oxidase can remain in the sol gel matrix. According to the consumption percentage of glucose, among four sets of samples ( I ) volume ratio of sol gel and ethanol 1:2 (II) volume ratio of sol gel and ethanol 1:4 (III) pH=1.5 (N) pH=lO, the highest glucose consumption is ( II) with 43.05%, followed by ( II) and (N) reaching 39.43% and 23.95%, respectively. The lowest activity of glucose oxidase is sol gel with pH=1.5, which can only oxidize 18.5% of glucose.
