Abstract: Acne Vulgaris is a common chronic skin inflammatory disorder caused by inflammation of the hair follicles and sebaceous glands of the skin, affecting millions of individuals worldwide. The presence of Priopionibacterium acnes (P. acnes) has been implicated as a major feature of Acne Vulgaris by its induction and maintenance of inflammatory conditions. Using a murine model of P. acnes infection, we characterise the contribution of lymphoid cells in the immune response to P. acnes and assess the role of Langerin-expressing DCs in the regulation of skin infiltrating immune cell recruitment. Our data show that in response to P. acnes, the αβ T and B lymphocytes expand in the skin-draining lymph node, suggesting a major activation of the immune response. This activation is translated by accumulation of T lymphocytes and ILCs in the skin. The depletion of Langerinexpressing DCs, which reportedly controls the innate immune response to P. acnes, significantly reduced cellularities in the skin-draining lymph node. However, the accumulation of T cells and ILCs in the skin is unaffected by the ablation of Langerin-expressing DCs.
