Title: Relationship between the level of haptoglobin and diabetic vascular complications

Abstract: A hemodynamic microfluidic chip system was implemented with the combination of caspase-3-based FRET biosensor cell line with microfluidic chip system which allows real time endothelial cell apoptosis. We introduced patient’s plasma samples into the microfluidic system with pulsatile flow profile, fixed glucose level and size of microchannel adjusted, to mimic the microenvironment of a blood vessel. An indication of vascular complications is endothelium dysfunction, which is caused by cell death through various complex biochemical pathways and Hp2-2. This study allows us to elucidate the combinational effects of physical, chemical and biological factors in the plasma. We investigated how different Hp genotypes affect endothelial cell apoptosis, which possibly lead to vascular complications. The results shows that pulsatile shear stress, level of glucose and plasma component play a role in endothelial cell death. Patients with Hp 2-2 exhibit a higher correlation compared to non Hp 2-2 group where apoptotic rate was compared with Hp level. These findings may provide new insights on the correlation of Hp 2-2 causing cell death and be used to develop an early detection method for diabetes vascular complication using the microfluidic technology based on the haptoglobin level.
